Luis Suarez is set to make his Uruguay return after a little less than two years away because of suspension — just in time to take on one of CONMEBOL’s traditional powerhouses in a match with plenty of importance.

World Cup qualifying in South America is set to resume on Thursday with the start of the fifth round of the campaign, but the one game many observers are most eager to watch takes place on Friday, when Uruguay travels to face Brazil. The battle between the two nations was already compelling because of where they each stand in the qualifying table, but Suarez playing in his first game since the 2014 World Cup only adds to the intrigue.

The prolific striker has been suspended from international duty since that tournament because of his infamous biting episode with Italy’s Giorgio Chiellini, but makes his way back to Uruguay in arguably the best form of his career and with an eagerness that spells trouble for Brazil.

Suarez’s club teammate, Leo Messi, will also be back in action for his respective country. Messi missed Argentina’s first four qualifiers of the cycle because of injury, but is ready to try and help his nation prevail in a tough road tilt against Chile.

The other three matches of Matchday 5 also contain interesting storylines and will provide a glimpse of what to expect in this summer’s Copa America Centenario, as Ecuador welcomes Paraguay, Peru locks horns with Venezuela and Colombia takes on Bolivia.

Here is a closer look at the upcoming fifth round of CONMEBOL World Cup qualifying:

BOLIVIA vs. COLOMBIA
(Thursday: 4 p.m. – beIN Sports Espanol, beIN Sports Connect)

Colombia is in need of points after picking up just four in as many games, but has the difficult challenge of having to play on the road in Bolivia’s altitude. The good news for Los Cafeteros is they have a full stable of attacking weapons available, including captain James Rodriguez, Juan Cuadrado, Adrian Ramos and Carlos Bacca.

Talent does not automatically mean production, however. Colombia has struggled to find the back of the net thus far in qualifying, netting just three times and looking little like the exciting team that generated a good share of chances at the most recent World Cup.

That is good news for Bolivia, which has been far from solid defensively. La Verde has yet to post a shutout, and has in fact given up two goals in every match so far. Being at home inside the comfort of Estadio Hernando Siles in La Paz should help, but players like defender and captain Edward Zenteno will have to step up in order to get the job done.

ECUADOR vs. PARAGUAY
(Thursday: 5 p.m. – beIN Sports, beIN Sports Connect)

The surprise of the campaign so far, first-placed Ecuador enters this home game at Estadio Olimpico Atahualpa in Quito as the only perfect nation remaining. La Tricolor is flawless at 4-0-0 thus far, and has looked pretty convincing at times with its combination of speed, physicality and technical skills.

Ecuador’s high-scoring attack has been a key ingredient to the early success, but Paraguay’s back line might prove tough to breakdown after conceding just twice so far. Still, the responsibility of creating chances and finding the necessary goals will fall on the shoulders of Antonio Valencia and Jaime Ayovi.

On the other side, Paraguay will aim to find its scoring touch. Los Guaranies have netted just three times, and know they need to do better in the attack given they are facing a strong Ecuadorian defense at altitude. Seattle Sounders forward Nelson Haedo Valdez will not be able to help in that regard as he was not summoned for this round of qualifiers, but Malaga’s Roque Santa Cruz and Edgar Benitez of Queretaro were and will have to carry the load.

CHILE vs. ARGENTINA
(Thursday: 7:30 p.m. ET – beIN Sports, beIN Sports Espanol, beIN Sports Connect)

The rematch of the 2015 Copa America final comes with plenty of pressure for both sides. Chile and Argentina have each had a tough time trying to play to their best, but could make a good jump up the table with a win at Estadio Nacional in Santiago.

Chile currently sits in fifth place with seven points, but has failed to win each of its last two qualifiers. La Roja most recently suffered a 3-0 humbling at the hands of Uruguay, and prior to that settled for a 1-1 draw at home against Colombia. A win is needed to stave off criticism and a dip in the standings, but a tough task lies ahead given that midfield ace Arturo Vidal, goal-scorer Eduardo Vargas and playmaker Jorge Valdivia are all unavailable because of suspension.

Making things even more difficult for Chile is the fact that Messi should be on the field for his first qualifier of the cycle. Messi was absent from the 2015 games because of an injury, but now returns to provide a boost to an Argentina side that has been offensively anemic without him. La Albiceleste has scored just twice so far to stand uncomfortably in sixth place, and could certainly use some Messi magic in Chile.

PERU vs. VENEZUELA
(Thursday: 10:15 p.m. – beIN Sports, beIN Sports Espanol, beIN Sports Connect)

It might be too early to call this a must-win game for Peru and Venezuela, but both of the basement-dwelling nations are acting like it is.

Peru and Venezuela have gotten off to tepid starts in qualifying, and know another poor result would put them in a bigger hole that might be too tough to climb out of. The ninth-placed Blanquirroja have the pressure of needing to win at home at Estadio Nacional in Lima, but Venezuela is in need of a result, preferably a victory, after failing to pick up any points thus far.

One key matchup to watch here is Claudio Pizarro against Oswaldo Vizcarrondo. The Peruvian striker has been red-hot as of late for Werder Bremen, and Vizcarrondo will have to do a good job of not allowing Pizarro too much space in the penalty area if La Vinotinto are to have a shot at pulling off the upset.

BRAZIL vs. URUGUAY
(Friday: 8:45 p.m. – beIN Sports Connect, beIN Sports Espanol)

The two nations closed out 2015 with lopsided 3-0 wins, and will be hoping to carry the momentum over into this first showdown of the year.

Uruguay will be on the road here, visiting the Itaipava Arena Pernambuco in Recife, but will welcome back top striker Suarez. Suspended coincidentally since the 2014 World Cup in Brazil, Suarez is hungry to deliver for Los Charruas after seeing them play so well without him. Uruguay is in second place now, and its only blemish through four matches was a narrow loss to table-leading Ecuador.

Brazil, meanwhile, is aiming to keep its unbeaten streak going. O Canarinho have not tasted defeat in their last three matches, and will be counting heavily on Neymar to keep things going in the attacking end. Brazil has scored seven times during this good run of form, but Uruguay’s stingy and physical defense figures to provide a stiff test even without the injured Diego Godin.
